ACUTE TOXICITY TEST OF DIMETHYL-4, 4¢¥-DIMETHOXY-5 6,5¢¥,6¢¥-DIMETHYLENE DIOXYBIPHENYL-2, 2¢¥-DICARBOXYLATE IN COMBINATION WITH GARLIC OIL IN RATS


Abstract
Previous studies have shown that dime¡©thyl-4, 4¢¥-dimethoxy-5,6,5 ,6¢¥-dimethylenedioxybiphenyl¡©2,2¢¥-dicarboxylate (PMC) in combination with garlic oil was effective in protecting (and/or treating) chemical-induced hepatic injury and that treatment of animals with the fo¡©rmulation synergistically improved chemical-induced im¡©pairment of hepatic triglycerides and cholesterol contents. In this study, the acute toxicity test was performed with the formulation of PMC and garlic oil in order to examine whether notable toxicities exist following a single large dose of both PMC and garlic oil administration. Treatment of male Sprague-Dawley rats with a single dose of the mixture containing PMC and garlic oil at the ratio of 1:4, resulted in no mortality in the entire animal groups exa¡©mined at the various doses from 0.5 g/kg through 8 g/kg body weight. No mortality was observed in female rats following treatment with the agents, representing that the mixture exhibited no gender-related toxicity. No clinical manifestations of acute toxicity were noted except dec¡©reased activities at the high doses : although animal ac¡©tivities were decreased at the dose of 8 g/kg for the first 6 hours and to a less extent at the dose of 4 g/kg in both sexes, animals resumed normal activity in 24 hours, in
